L’imagerie et la spectroscopie par résonance magnétique nucléaire (RMN) sont des techniques puissantes permettant d’étudier in vivo l’anatomie, la physiologie et la biochimie du muscle au cours d’une unique séance d’examens.
En outre, le contraste intrinsèque du muscle par RMN est extrêmement riche et des séquences d’impulsions radiofréquences adaptées permettent de quantifier spécifiquement différentes variables comme la fraction de graisse intramusculaire, les temps de relaxation de l’eau musculaire, la perfusion ou l’oxygénation du muscle, qui sont potentiellement altérées chez des sujets atteints de pathologies neuromusculaires.
Le laboratoire concentre la plupart de ses efforts sur le développement de ces méthodes RMN quantitatives, leur validation, leur promotion, et leur utilisation dans le cadre d’essais cliniques mono-centriques et multi-centriques concernant les pathologies neuromusculaires. Ceux-ci visent à étudier l’histoire naturelle de ces maladies ou à évaluer l’effet de thérapies innovantes.
Plus précisément, nos projets de recherche s’intéressent :
- Au développement préclinique et clinique de nouveaux contrastes RMN permettant de détecter et de quantifier différents processus pathologiques impliqués dans l’apparition et la progression des myopathies, comme l’inflammation, la nécrose, et la fibrose musculaire.
- A déterminer la valeur prédictive des nouveaux contrastes RMN sur la progression de certaines pathologies.
- Au développement d’outils permettant d’étudier le muscle à l’exercice (ergomètres non-magnétiques, antennes radiofréquences, séquences RMN entrelacées, …).
- A l’accélération des acquisitions RMN grâce à l’implémentation de nouveaux schémas d’encodage du signal (acquisitions radiales sous échantillonnées) et la quantification simultanée de plusieurs variables (« fingerprinting » par RMN, méthodes d’apprentissage profond, …).
